Practical cautions

Use consistent units and assay conditions. Temperature, pH, ionic strength, enzyme concentration, cofactors, inhibitors, and substrate depletion can all change observed kinetics. Vmax normally changes with total enzyme concentration, while Km may shift when the mechanism or assay environment changes.
